Culture Medium:DMEM:Ham’s F12 media supplemented with 2 mM glutamine, 15 mM HEPES and 5% fetal bovine serum.
Subculturing:Remove medium of sub-confluent cultures and rinse with 0.02% EDTA solution. Add fresh 0.025% trypsin / 0.02% EDTA solution and let the culture sit at 37°C until cells detach. Add fresh medium, centrifuge and dispense into new flasks.
Split Ratio:A ratio of 1:3 to 1:8 is recommended
Fluid Renewal:Every 2 to 3 days.
Freeze Medium:CM-1 (Cell Lines Service)
Sterility:Tests for mycoplasma, bacteria and fungi were negative
Antigen Expression:HLA DR (on approx. 25% of cells)
