| Culture Medium: | Minimum essential medium Eagle supplemented with 2 mM L-glutamine and Earle's BSS adjusted to contain 1.5 g/L sodium bicarbonate, 0.1 mM non-essential amino acids, 1.0 mM sodium pyruvate and 10% heat-inactivated fetal bovine serum. Alternatively, DMEM:Ham's F12 medium supplemented with 2 mM L-glutamine and 10% fetal bovine serum may be used. |
| Subculturing: | Remove medium, add fresh 0.025% trypsin, 0.02% EDTA solution, rinse, remove trypsin, EDTA and allow the cultures to sit at room temperature (or 37°C for a short time period) until the cells detach. Add fresh medium, aspirate and dispense into new flasks. |
