Designation:SW 872
Depositor:Leibovitz
Organism:Homo sapiens (human)
Ethnicity:Caucasian
Age/Stage:36 years
Gender:male
Tissue:liposarcoma
Morphology:fibroblast
Growth Properties:monolayer
Description:The SW 872 cell line was initiated by A. Leibovitz in 1974 at the Scott and White Clinic, Temple, Texas from a surgical specimen of a fibrosarcoma removed from a 36 year old male Caucasian. The histopathology evaluation reported an undifferentiated malignant tumor consistent with liposarcoma.
Culture Medium:Leibovitz's L-15 medium supplemented with 2 mM L-glutamine and 5% fetal bovine serum. Alternatively, the cells may be cultured in DMEM, 4.5 g/L glucose, supplemented with 2 mM L-glutamine and 5% fetal bovine serum. (37°C, 5% CO2)
Subculturing:Remove medium, rinse with fresh 0.025% trypsin, 0.02% EDTA solution and let the culture sit at room temperature for two minutes. Remove trypsin, and let the culture sit at 37°C for 5 minutes. Add fresh medium to disperse the cells, and dispense into new flasks.
Split Ratio:A ratio of 1:3 to 1:6 is recommended
Fluid Renewal:2 to 3 times weekly
Freeze Medium:CM-1 (CLS ∙ Cell Lines Service)
Sterility:Tests for mycoplasma, bacteria and fungi were negative
Biosafety Level:1
Tumorigenic:yes, produces spindle cell sarcoma in nude mice consistent with liposarcoma
Karyotype:hypertriploid; modal number = 80; range = 66 to 81. The rate of higher ploidies was 8.2%. Ten markers were common to most cells. These were: der(5)t(5;?)(q31;?)1, der(5)t(5;?)(q31;?)2, der(6)t(6;?)(q15;?), der(7)t(7;?)(q36;?), t(15q16q) and five others. Both der(5) markers, the der(7) and t(15q16q) were paired. There were 5 copies of N20 and N21, 4 copies of N8, N9, N11, N14 and N17 and a single copy of X in each cell.
Antigen Expression:Blood type O+
Isoenzymes:G6PD, B; PGM1, 1-2; PGM3, 1; ES-D, 1; AK-1, 1; GLO-1, 1-2; Phenotype Frequency Product: 0.0708
