Description:HOS cells exhibit a flat morphology, low saturation density, low plating efficiency in soft agar and are sensitive to chemical and viral transformation. The cells express alkaline phosphatase under basal conditions (Clover J, 1994).
Culture Medium:Minimum essential medium (Eagle) in Earle's BSS supplemented with 2 mM L-glutamine, 1 mM non-essential amino acids (NEA) and 10% fetal bovine serum.
Subculturing:Remove medium and rinse with 0.02% EDTA solution. Add fresh 0.025% trypsin solution for 2 to 4 minutes at 37°C. Stop the enyme activity by adding fresh medium, aspirate and dispense into new flasks.
Split Ratio:A ratio of 1:2 to 1:4 is recommended
Fluid Renewal:2 to 3 times weekly
