Description:The SW 1353 cell line was initiated by A. Leibovitz at the Scott and White Clinic, Temple, Texas in 1977 from a primary grade II chondrosarcoma of the right humerus obtained from a 72 year old female Caucasian. The initial culture medium was L-15 containing cortisone and insulin plus 10% fetal bovine serum and antibiotics.
Culture Medium:Leibovitz's L-15 medium supplemented with 1% L-glutamine and 10% fetal bovine serum
Subculturing:Remove medium, rinse with fresh 0.025% trypsin solution, remove trypsin and let the culture sit at 37°C until the cells detach. Add fresh medium, aspirate and dispense into new flasks.
Split Ratio:A ratio of 1:2 is recommended
Fluid Renewal:2 to 3 times weekly
Freeze Medium:CM-1 (CLS ∙ Cell Lines Service)
Sterility:Tests for mycoplasma, bacteria and fungi were negative
