Description:This breast tumor line was established by E.Y. Lasfargues and L. Ozzello in 1958 by isolation and cultivation of cells spilling out of the tumor when it was cut in thin slices. A mycoplasma contaminant was discovered and eliminated early in 1972. Growth of BT-20 cells is inhibited by tumor necrosis factor alpha (TNF alpha). BT-20 cells are negative for estrogen receptor, but do express an estrogen receptor mRNA that has deletion of exon 5.
Culture Medium:DMEM:Ham's F12 mixture (1:1) supplemented with 2 mM L-glutamine and 10% fetal bovine serum
Subculturing:Remove medium and rinse with 0.02% EDTA (versene) solution. Add fresh 0.025% trypsin/0.02% EDTA (versene) solution, remove and let the culture sit at 37oC until the cells detach. Add fresh medium, remove trypsin by centrifugation and dispense into new flasks. Subculture every 6 to 8 days.
