Description:The BT-549 line was isolated in 1978 by W.G. Coutinho and E.Y. Lasfargues. Source tissue consisted of a papillary, invasive ductal tumor which had metastasized to 3 of 7 regional lymph nodes. The established population was polymorphic with epithelial like components and multinucleated giant cells. A mucin-like material was secreted into the medium.
Culture Medium:RPMI 1640 medium with 0.023 IU/ml insulin, 90%; fetal bovine serum, 10%
Subculturing:Remove medium, add fresh 0.025% trypsin, 0.02% EDTA solution, swirl gently, remove trypsin and let the culture sit at 37°C for 3 to 5 minutes. Add fresh medium, aspirate and dispense into new flasks. When cultures become confluent some cells will slough off into the medium, these cells can be centrifuged and placed into new culture flasks.
