Description:MDA-MB-435S is a spindle shaped strain which evolved from the parent line (435) isolated in 1976 (R. Cailleau et al.) from the pleural effusion of a 31 year old female with metastatic ductal adenocarcinoma of the breast. Parental cells displayed the dispersed pattern (type II) when treated with immunofluorescent stain for tubulin. Recent findings point to a rather melanocytic origin of this cell line (Ellison 2002).
Culture Medium:DMEM:Ham's F12 supplemented with 2 mM L-glutamin and 10% fetal bovine serum. Incubate at 37°C/5%CO2.
Subculturing:Remove medium and rinse with fresh 0.02% EDTA solution. Add fresh trypsin (0.025%)/EDTA (0.03%) solution, rinse and remove. Place flask at 37°C until the cells detach. Add fresh medium, carefully resuspend the cells and dispense into new flasks.
Split Ratio:A ratio of 1:3 to 1:6 is recommended
