Designation:LS-174T
Depositor:CLS
Organism:Homo sapiens (human)
Ethnicity:caucasian
Age/Stage:58 years; Duke's type B
Gender:female
Tissue:Colon, colorectal
Morphology:epithelial
Celltype:adenocarcinoma
Description:The cells are positive for Colon Antigen 3. They are negative for p53 antigen expression, but positive for mRNA expression. The line was derived from the same tumor as LS 180. LS 174T cells stain positively for cytokeratins.
Culture Medium:Minimum essential medium (Eagle) in Earle's BSS supplemented with 2 mM L-glutamin, 1% non-essential amino acids and 10% fetal bovine serum.
Subculturing:Remove medium and rinse with fresh 0.02% EDTA solution. Add fresh 0.025% trypsin/0.02 M EDTA and let the culture sit at 37°C until the cells detach. Add fresh cell growth medium, remove trypsin by centrifugation, resuspend the cells in fresh culture medium and dispense into new flasks.
Split Ratio:A ratio of 1:2 to 1:5 is recommended
Fluid Renewal:2 to 3 times weekly
Freeze Medium:CM-1 (CLS ∙ Cell Lines Service)
Sterility:Tests for mycoplasma, bacteria and fungi were negative
Biosafety Level:1
Tumorigenic:yes, in nude mice
Oncogene:myc +; myb + ; ras +; fos +; p53 +; sis -; abl -; ros -; src -
Karyotype:45,X; one X chromosome missing; no other chromosomal aberrations
