Designation:MEWO
Depositor:CLS
Organism:Homo sapiens (human)
Ethnicity:Caucasian
Tissue:melanoma, malignant; skin
Morphology:stellate
Growth Properties:adherent
Description:Product melanin, derived from a human melanoma by Prof.C.Grose (1978). The cells support growth of varicella-zoster virus (VZV) isolates at 36°C although growth is optimal at 32°C.
Culture Medium:Minimum essential medium (Eagle) in Earle's BSS supplemented with 2 mM L-glutamine, 1% nonessential amino acids, 1 mM sodium pyruvate and 10% fetal bovine serum.
Alternatively, DMEM:F12 (1:1 mixture) supplemented with 2 mM L-glutamine and 10% fetal bovine serum may be used.
Subculturing:Remove medium, add fresh 0.025% trypsin solution for 1 minute, remove trypsin and let the culture sit at room temperature for 5 to 10 minutes. Add fresh medium, aspirate and dispense into new flasks. Subculture every 6 to 8 days.
Split Ratio:A ratio of 1:3 to 1:6 is recommended
Fluid Renewal:2 to 3 times per week
Freeze Medium:CM-1 (CLS ∙ Cell Lines Service)
Sterility:Tests for mycoplasma, bacteria and fungi were negative
Biosafety Level:1
Tumorigenic:yes, in nude mice; forms malignant melanoma
DNA Profile (STR):Amelogenin: X / Y
