| Depositor: | CLS |
| Organism: | Homo sapiens (human) |
| Ethnicity: | Caucasian |
| Tissue: | melanoma, malignant; skin |
| Morphology: | stellate |
| Growth Properties: | adherent |
| Description: | Product melanin, derived from a human melanoma by Prof.C.Grose (1978). The cells support growth of varicella-zoster virus (VZV) isolates at 36°C although growth is optimal at 32°C. |
| Culture Medium: | Minimum essential medium (Eagle) in Earle's BSS supplemented with 2 mM L-glutamine, 1% nonessential amino acids, 1 mM sodium pyruvate and 10% fetal bovine serum. Alternatively, DMEM:F12 (1:1 mixture) supplemented with 2 mM L-glutamine and 10% fetal bovine serum may be used. |
| Subculturing: | Remove medium, add fresh 0.025% trypsin solution for 1 minute, remove trypsin and let the culture sit at room temperature for 5 to 10 minutes. Add fresh medium, aspirate and dispense into new flasks. Subculture every 6 to 8 days. |
| Split Ratio: | A ratio of 1:3 to 1:6 is recommended |
| Fluid Renewal: | 2 to 3 times per week |
| Freeze Medium: | CM-1 (CLS ∙ Cell Lines Service) |
| Sterility: | Tests for mycoplasma, bacteria and fungi were negative |
| Biosafety Level: | 1 |
| Tumorigenic: | yes, in nude mice; forms malignant melanoma |
| DNA Profile (STR): | Amelogenin: X / Y CSF1PO: 12/12 D13S317: 8/9 D16S539: 10/12 D5S818: 12/13 D7S820: 10/12 THO1: 7/9 TPOX: 8/10 vWA: 15/15 D3S1358: 17/17 D21S11: 30/32.2 D18S51: 14/17 Penta E: 5/5 Penta D: 10/10 D8S1179: 13/15 FGA: 22/22 CLS ∙ Cell Lines Service, 2009. |
