Designation:A-427
Depositor:CLS
Organism:Homo sapiens (human)
Ethnicity:Caucasian
Age/Stage:52 years
Gender:male
Tissue:lung
Morphology:epithelial
Celltype:carcinoma
Growth Properties:monolayer
Description:The A-427 cell line was established by D.J. Giard in 1973.
Culture Medium:DMEM: Ham's F12 medium (1:1, vol:vol) supplemented with 2 mM L-glutamine and 10% fetal bovine serum.
Subculturing:Remove medium and rinse with fresh 0.02% EDTA solution. Add 0.025% trypsin/0.02% EDTA solution and let the culture sit at 37°C until the cells detach. Add fresh medium, centrifuge to remove trypsin, aspirate and dispense into new flasks.
Split Ratio:A ratio of 1:2 to 1:6 is recommended
Fluid Renewal:2 times weekly
Freeze Medium:CM-1 (CLS ∙ Cell Lines Service)
Sterility:Tests for mycoplasma, bacteria and fungi were negative
Tumorigenic:yes, in nude mice; forms an undifferentiated tumor suggestive of adenocarcinoma
Karyotype:(P60) hypotriploid to hypertriploid with abnormalities including dicentrics, minutes and large subtelocentric marker
Isoenzymes:PGM3, 1; PGM1, 1-2, ES-D, 1; AK-1, 2; GLO-1, 1; G6PD, B; Phenotype Frequency Product: 0.00006
