| Designation: | Capan-1 |
| Organism: | Homo sapiens (human) |
| Ethnicity: | Caucasian |
| Age/Stage: | 40 years |
| Gender: | male |
| Tissue: | Pancreas (from metastatic site: liver) |
| Morphology: | epithelial |
| Celltype: | adenocarcinoma |
| Growth Properties: | monolayer |
| Description: | The cells will slough off of the growth surface if they become too heavy. Capan-1 expresses the cystic fibrosis transmembrane conductance regulator (CFTR) and secretes gastric type mucin. |
| Culture Medium: | RPMI 1640 medium supplemented with 2 mM Glutamine and 10% foetal bovine serum. |
| Subculturing: | Remove medium and rinse with EDTA (versene) solution. Add fresh 0.025% trypsin / EDTA (versene) solution and let the culture sit at 37°C until the cells detach. Add fresh medium, centrifuge to remove trypsin, resuspend in fresh medium and dispense into new flasks. Subculture every 6 to 8 days. |
| Split Ratio: | A ratio of 1:2 to 1:4 is recommended |
| Fluid Renewal: | 2 times weekly |
| Freeze Medium: | CM-1 (CLS ∙ Cell Lines Service) |
| Sterility: | Tests for mycoplasma, bacteria and fungi were negative |
| Biosafety Level: | 1 |
| Tumorigenic: | yes, in nude mice; forms adenocarcinoma consistent with pancreatic duct carcinoma |
| Karyotype: | (P7) hypotriploid with abnormalities including dicentrics, breaks, acrocentric fragments, large submetacentric and subtelocentric chromosomes plus minute marker |
| DNA Profile (STR): | Amelogenin: X,X CSF1PO: 11 D13S317: 9 D16S539: 13,14 D5S818: 11 D7S820: 10,11 THO1: 6 TPOX: 8,11 vWA: 16 D3S1358: 15 D21S11: 28,30 D18S51: 12 Penta E: 10,12 Penta D: 9,13 D8S1179: 14,16 FGA: 24 CLS ∙ Cell Lines Service, 2010. |
