Designation:Capan-1
Organism:Homo sapiens (human)
Ethnicity:Caucasian
Age/Stage:40 years
Gender:male
Tissue:Pancreas (from metastatic site: liver)
Morphology:epithelial
Celltype:adenocarcinoma
Growth Properties:monolayer
Description:The cells will slough off of the growth surface if they become too heavy. Capan-1 expresses the cystic fibrosis transmembrane conductance regulator (CFTR) and secretes gastric type mucin.
Culture Medium:RPMI 1640 medium supplemented with 2 mM Glutamine and 10% foetal bovine serum.
Subculturing:Remove medium and rinse with EDTA (versene) solution. Add fresh 0.025% trypsin / EDTA (versene) solution and let the culture sit at 37°C until the cells detach. Add fresh medium, centrifuge to remove trypsin, resuspend in fresh medium and dispense into new flasks. Subculture every 6 to 8 days.
Split Ratio:A ratio of 1:2 to 1:4 is recommended
Fluid Renewal:2 times weekly
Freeze Medium:CM-1 (CLS ∙ Cell Lines Service)
Sterility:Tests for mycoplasma, bacteria and fungi were negative
Biosafety Level:1
Tumorigenic:yes, in nude mice; forms adenocarcinoma consistent with pancreatic duct carcinoma
Karyotype:(P7) hypotriploid with abnormalities including dicentrics, breaks, acrocentric fragments, large submetacentric and subtelocentric chromosomes plus minute marker
DNA Profile (STR):Amelogenin: X,X
