Description:The cells are positive for keratin by immunoperoxidase staining.
Culture Medium:Minimum essential medium (Eagle) in Earle's BSS with non-essential amino acids, sodium pyruvate (1 mM) and lactalbumin hydrolysate (0.1%), 90%; fetal bovine serum, 10%. Alternatively DMEM:F12 (1:1 mixture) supplemented with 2 mM L-glutamine and 10 % fetal bovine serum may be used.
Subculturing:Remove medium, add fresh 0.025% trypsin for 2 to three minutes, remove trypsin and let the culture stand for 5 to stand for 5 to 10 minutes at room temperature. Add fresh medium, aspirate and dispense into new flasks.
Split Ratio:A ratio of 1:2 to 1:4 is recommended
Fluid Renewal:3 times weekly
