| Description: | The SVI cell line has been cloned from the outgrowth of glomeruli which were isolated from H-2kb-tsA58 transgenic mice. The mice carry a temperature-sensitive variant of the SV40 large T antigen under control of the IFN-g-inducible H-2kb promoter. Cells proliferate at 33°C, and they differentiate at 38°C. At present, the cells have been cultured successfully for more than 40 passages without noting phenotypic changes. |
| Culture Medium: | RPMI 1640 media supplemented with 2 mM L-glutamine and 10% fetal bovine serum |
| Subculturing: | Remove medium, wash with PBS; add fresh trypsin/versene solution (0.05% trypsin, 0.02% EDTA) for 3 minutes at 38°C. Add fresh medium, centrifuge and dispense into new flasks. |
| Split Ratio: | A ratio of 1:2 to 1:3 (38°C) or 1:5 (33°C) is recommended |
